Distribution characteristics of trace metals in wheat plants and response of phyllosphere microorganisms under fly ash stress

Deposition of fly ash (FA) emitted from coal-fired electricity has been recognized as an important trace metals source for crops growing in overlapped areas of farmland and coal resources. In this study, we investigated total content and chemical form distribution of the trace metals in wheat (Liangxing 99), and phyllosphere microbial community to discussed how phyllosphere microorganisms affect wheat uptake of trace metals from FA and metals migration and transformation in plant. Results showed that: (1) FA increased the content of Cu, Zn, and Pb in wheat shoot significantly, and also Cr and Pb in the root. (2) Cr, Zn, and Pb are mainly extracted with hydrochloric acid and residual in both shoot and root (over 60%), while Cu is mainly extracted with ethanol and deionized water (nearly 70%). (3) The addition of FA reduces the diversity of phyllosphere microorganisms and increase the richness of the microbial community. The dominant bacterial communities are Proteobacteria and Bacteroidetes. Characteristic bacterium is closely related to total and ethanol extracted metals in shoot. This study provides evidence for the trace metals tolerance and detoxification mechanisms of wheat at the levels of chemical morphology and leaf microbiota.
